Applications
Animals were immunized against exotoxin A of Pseudomonas aeruginosa by infection with strain PA220.
The antibody binds specifically for the exotoxin A of Pseudomonas aeruginosa.
It displays specificity for exotoxin A from strain PA103 and strain PAO1 and PAO-PR1, a derivative of PAO1.
The antibody shows high specificity and reactivity for native and denatured exotoxin A in ELISA assays.
Both TC1 (ATCC CRL-2493) and TC31 (ATCC CRL-2494) antibodies neutralize exotoxin A toxicity against CHO cells in an in vitro assay but only TC31 protected mice infected with strain PA220 in an in vivo model.
A hybridoma producing an anti idiotypic antibody against TC31 is available (ATCC CRL-2485).

Derivation
Spleen cells were fused with Sp2/0-Ag14 cells.

Subculturing
Protocol: Cultures can be maintained by addition of fresh medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 5 X 10(4) viable cells/ml. Do not allow the cell concentration to exceed 5 X 10(5) viable cells/ml.
Interval: Maintain cultures at a cell concentration between 5 X 10(4) and 5 X 10(5) cells/ml.
Medium Renewal: Add fresh medium every 2 to 3 days (depending on cell density)
Cryopreservation
Freeze medium: Complete growth medium supplemented with 5% (v/v) DMSO
