Comments
Animals were immunized with TC-31 (ATCC CRL-2494), a monoclonal antibody to exotoxin A of Pseudomonas aeruginosa.
Spleen cells were fused with Sp2/0-Ag14 myeloma cells.
The antibody binds specifically to an idiotypic determinant on TC-31 anti-exotoxin A monoclonal antibody. Exotoxin A inhibits the binding of the 9-2-4 antibody to TC-31.
This antibody conjugated to keyhole limpet hemocyanin (KLH) can induce antibodies in mice that bind to exotoxin A and protect 80% of the mice from a lethal injection of exotoxin A.

Subculturing
Protocol: Cultures can be maintained by addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension in fresh medium at 4 to 5 X 10(5) viable cells/ml. Do not allow the cell concentration to exceed 1 X 10(6) cells/ml.
Interval: Maintain cultures at a cell concentration between 5 X 10(5) and 1 X 10(6) cells/ml.
Medium Renewal: Add fresh medium every 2 to 3 days (depending on cell density)

Freeze medium: Complete growth medium supplemented with 5% (v/v) DMSO
