Animals were immunized human brain homogenate of multiple sclerosis plaques obtained at autopsy.
Spleen cells were fused with NS-1 myeloma cells.
The antibody recognizes a novel intermediate filament associated protein (MW 100000) with similarity to alpha actinin.
This antibody intensely stains astroglia and related macroglia in cryostat sections of human and rat central nervous system tissue.
It is useful for immunofluorescence, immunocytochemistry, and Western blotting procedures.
Complete Growth Medium The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
Subculturing
Protocol: Cultures can be maintained by addition of fresh medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 1 X 10(5) viable cells/ml. Do not allow the cell concentration to exceed 1 X 10(6) cells/ml.
Interval: Maintain cultures at a cell concentration between 0.5 X 10(5) and 1 X 10(6) cells/ml.
Medium Renewal: Every 2 to 3 days
Cryopreservation
Freeze medium: Complete growth medium supplemented with 5% (v/v) DMSO
Storage temperature: liquid nitrogen vapor phase
Culture Conditions
Atmosphere: air, 95%; carbon dioxide (CO2), 5%
Temperature: 37.0°C
