Derivation

Animals were immunized with purified integrin associated protein (which is physically and functionally associated with vitronectin receptors).

Spleen cells were fused with P3X63Ag8.653 myeloma cells.

Comments

The antibody reacts with vitronectin receptors on human polymorphonuclear leukocytes (PMN, neutrophils) and blocks the receptor binding to the tripeptide Arg-Gly-Asp (RGD).

The antibody also inhibits RGD stimulated phagocytosis.

It precipitates a heterodimer of apparent molecular weights 140000 and 105000 daltons.

The antibody does not react with gpIIb/III or the LFA-1/Mac-1 family of cellular adhesion receptors.

Subculturing Cultures can be maintained by the addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 2 x 10viable cells/mL. Maintain cultures at cell concentrations between 2 x 105 and 1 x 10viable cells/mL.
Medium Renewal: Every 2 to 3 days
Cryopreservation

Complete culture medium described above supplemented with 5% (v/v) DMSO.  Cell culture tested DMSO is available as ATCC Catalog No. 4-X.
