Organism Homo sapiens, human Tissue peritoneal effusion Morphology lymphoblast-like Culture Properties suspension, multicell aggregates Biosafety Level 1 Disease large cell lymphoma; B cell lymphoma Storage Conditions liquid nitrogen vapor phase Karyotype t(14;18) chromosomal translocation, positive Genes Expressed methionine aminopeptidase 2 (MetAP2), expressed cathelicidin antimicrobial peptide (CAMP) expression is induced upon exposure to 1,25(OH)2D3 bcl-6 protein, expressed Comments The depositor states that this cell line is negative for the presence of Epstein-Barr virus(EBV) genome. ATCC confirmed this cell line is negative for the presence of Epstein-Barr viral DNA sequences via PCR. The SU-DHL-16 cell line possesses a t(14;18)(q32;q21) translocation. The SU-DHL-16 cell line contains a minor bcl-2 rearrangement. Complete Growth Medium The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%. Subculturing Protocol: Cultures can be maintained by the addition of fresh medium. An inoculum of 1 X 105 to 2 x 105 cells/mL is recommended. Subculture when cell concentration is between 1 x 106 and 2 x 106 cells/mL. Subcultivation ratio: A subcultivation ratio of 1:4 to 1:10 is recommended. Medium renewal: Add fresh medium every 2 to 3 days (depending on cell density) Cryopreservation Freeze medium: complete growth medium supplemented with an additional 20% fetal bovine serum and 10% (v/v) DMSO Storage temperature: liquid nitrogen vapor phase Culture Conditions Temperature: 37.0°C Atmosphere: air, 95%; carbon dioxide (CO2), 5% STR Profile D5S818: 10 D13S317: 9, 12 D7S820: 9, 10 D16S539: 11, 13 vWA: 15, 18 THO1: 7, 9.3 TPOX: 8, 11 CSF1PO: 10, 11 Amelogenin: XY Name of Depositor A Epstein Year of Origin 1980 References Siminovitch KA, et al.
