Karyotype modal number = 53. This is a hypotriploid human cell line with the modal chromosome number occurring in 68% of cells counted. The polyploid cells occurred at 3.0%; eleven marker chromosomes were common to all cells, of which two were paired. The markers included: der(6)(q13), t(7p17q), del(10)(p11.23), paired der(7)t(7;?17)(p22;?q21.1) and der(16)t(16;?)(q24;?), and six others; normal Y, N13, N15, and N16 were absent; the X chromosomes were mostly paired.
Derivation
This line was derived by A.F. Gazdar, H.K. Oie, J.D. Minna and associates from a lung cancer obtained from a patient prior to therapy.­

Comments

The cells carry a mutation in codon 191 of the TP53 gene.

The cells are not able to synthesize the peptide neuromedin B (NMB) or the gastrin releasing peptide (GRP).

The cells stain positive for keratin and vimentin but are negative for neurofilament triplet protein.

Complete Growth Medium The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
Subculturing

Remove medium, and rinse with 0.25% trypsin, 0.53 mM EDTA solution. Remove the solution and add an additional 1 to 2 mL of trypsin-EDTA solution. Allow the flask to sit at room temperature (or at 37°C) until the cells detach. Add fresh culture medium, aspirate and dispense into new culture flasks.

Subcultivation Ratio: A subcultivation ratio of 1:3 to 1:6 is recommended
