Derivation
Animals were immunized with recombinant human thyroid stimulating hormone (TSH) receptor expressed in. E. coli. Spleen cells were fused with Sp2/0 mouse myeloma cells. The antibody recognizes the N-terminal extracellular domain (domain amino acids 19-246), alpha subunit of the human thyroid stimulating hormone (TSH) receptor (hormone binding unit). The TSHR-T5-51 (ATCC CRL-2680), TSHR-R5T-44 (ATCC CRL-2681) and TSHR-T5U-317 (ATCC CRL-2682) antibodies are additive. They recognize different epitopes. TSHR-T5U-317 cross reacts with the rodent TSHR The antibodies do not interfere with hormone binding and have no effect on the receptor mediated adenylate cyclase activity. Complete Growth Medium The base medium for this cell line is ATCC-formulated Dulbecco's Modified Eagle's Medium, Catalog No. 30-2002. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
Subculturing
Protocol: Cultures can be maintained by the addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 3 to 5 X 10(4) viable cells/ml.
Interval: Maintain cell density between 3 X 10(4) and 4 X 10(5) viable cells/ml.
Medium Renewal: Add fresh medium every 2 to 3 days (depending on cell density)

Freeze medium: Complete growth medium supplemented with 5% (v/v) DMSO
