Derivation
The HMy2.CIR cell line was derived from the HMy.2 B lymphoblastoid cell line, a fast growing mutant of the ARH-77 cell line (ATCC CRL-1621), by gamma irradiation and selection for loss of HLA class I antigen expression.
Comments

The CRL-1621cell line is reported to be positive for Epstein-Barr nuclear antigen (EBNA +) and Epstein-Barr viral capsid antigen (EBVCA +). 

Since the HMy2.CIR cell line was derived from the HMy.2 B lymphoblastoid cell line, a fast growing mutant of the ARH-77 cell line (ATCC CRL-1621), it is assumed to be EBNA positive.

The cells express no HLA A or B locus products, but do express small amounts of HLA Cw4.


Complete Growth Medium The base medium for this cell line is ATCC-formulated Iscove's Modified Dulbecco's Medium, Catalog No. 30-2005. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
Subculturing
Cultures can be maintained by addition or replacement of fresh medium. Start cultures at 2 X 105 viable cells/mL.
Interval: Maintain between 2 X 105 and 1 X 106 viable cells/mL.
Medium Renewal: Every 2 to 3 days
Cryopreservation
Freeze medium: Complete growth medium supplemented with 5% (v/v) DMSO
Storage temperature: liquid nitrogen vapor phase
