Complete Growth Medium RPMI 1640 with 2.0 mM L-glutamine adjusted to contain 1.5 g/L sodium bicarbonate, 4.5 g/L glucose, 10 mM HEPES, and 1.0 mM sodium pyruvate, 80%; fetal bovine serum, 10%; rat T-STIM with Con A, 10%. Rat T-STIM with Con A can be obtained from Becton Dickinson, Catalogue No. 354115
Subculturing
Medium Renewal: Every 2 days
Cultures can be maintained by addition or replacement of fresh medium. Start cultures at 1 X 10 exp5 cells/ml and maintain between 0.5 X 10 exp5 and 3 X 10 exp6 cells/ml.
