Karyotype Modal chromosome number: 74. This is a hypertriploid male cell line, del(6)(q16), t(8;11)(q22;q23), add(11)(q25), add(13)(q34), add(14)(q32). Most cells contain the t(11;14) derivative chromosome, detected by FISH with whole chromosome paint probes.

Comments
The cells are large, growing singly and in small clumps in vitro. The immunophenotype by flow cytometry is compatible with MCL. Western blots show expression of cyclin D1 but no detectable cyclin D2 and cyclin D3; the retinoblastoma protein is predominantly phosphorylated. There is expression of tumor suppressor gene products including p53, p16 (INK4a), and p21(WAF1).
Complete Growth Medium The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 15%.
Subculturing

Cultures can be maintained by the addition of fresh medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 6 X 104 viable cells/mL. Do not allow the cell concentration to reach 1.5 X 106 cells/mL.
