Restriction digests of the clone give the following sizes (kb): BamHI--2.9, EcoRI--2.9, PstI--2.9. Cloning into the BamHI site followed by transformation into an EcoK r+ m+ host, increases the relative transformation efficiency of recombinant molecules, provided that the cloned insert is free of non-methylated EcoK sites. Vector allowing positive selection and blue/white color detection for cloned inserts. Vector constructed from pTZ19R by elimination of the EcoK restriction site in the ampR region and creation of a new EcoK site within the polylinker, containing a BamHI site as the central hexamer of the EcoK recognition sequence.
