Excellent preparation, low noise, high resolution. Expect purity above 90-95%. Over 80% of clones characterized from this library mapped to chromosome 22; one of 10 mapped clones mapped to an inappropriate chromosome. Of 493 inserts analyzed, 179 were single-copy specific to chromosome 22, 16 were single copy on 22 but detected other loci, 134 were single copy not on chromosome 22, and 164 were difficult to analyze. The value listed under genome equivalents is for the amplified library. Coverage was calculated by dividing the number of independent recombinants by the genome equivalents of the unamplified library. This value is 71. Of 384 random clones, 199 did not hybridize to total human DNA. Of 154 clones analyzed, 79 had inserts larger than 0.6 kb. Of these, 39 were single copy, with 15 mapping to 22, 4 mapping to 21, and 20 mapping to neither. Of 195 single copy probes analyzed, the distribution among 4 regions of the chromosome was proportional to the estimated genetic length of these regions, suggesting random representation in the library.
