Cytogenetic Analysis: modal number = 69; range = 65 to 74.
This is a pseudotriploid human cell line with the modal chromosome number of 69, occurring in 24% of cells. However, cells having chromosome counts from 65 to 70 also occurred at high frequencies. The rate of cells with higher ploidies was 7%. There were eight marker chromosomes common to most cells. They included paired del(1) (p22), t(8q11q), i(14q), single i(7p) and four others. Two more markers were seen only in some cells. Only one normal X chromosome was found in G-banded preparation. The Y chromosome was not detected in the Q-banded preparation.

Comments: The cells produce high levels of MUC-1 mucin mRNA, low levels of MUC-2 mRNA but do not express the MUC-3 gene.
Propagation: ATCC complete growth medium: The base medium for this cell line is ATCC-formulated McCoy's 5a Medium Modified, Catalog No. 30-2007. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.

Subculturing: Protocol: Remove medium, and rinse with 0.25% trypsin, 0.53 mM EDTA solution. Remove the solution and add an additional 1 to 2 ml of trypsin-EDTA solution. Allow the flask to sit at room temperature (or at 37C) until the cells detach.Add fresh culture medium, aspirate and dispense into new culture flasks.
Subcultivation Ratio: A subcultivation ratio of 1:2 to 1:4 is recommended
