Comments: The cell line is cross-resistant to anthracycline analogues including daunomycin, epirubicin, menogaril, and mitoxantrone as well as to acivicin, etoposide, gramicidin D, colchicine, and the Vinca alkaloids, vincristine and vinblastine. [49663]
The cells display little or no cross-resistance to bleomycin, 5-fluorouracil, and carboplatin. They have a slight collateral sensitivity to 1-dehydrotestosterone and lidocaine. [49663]
The H69AR cell line is approximately 50-fold resistant to adriamycin as compared to the parental NCI-H69 cell line. [49655]
The monoclonal antibodies QCRL-1 (ATCC HB-11765) and QCRL-3 (ATCC HB-11766) will react with fixed H69AR cells. [49661]
The antibodies reacts with MRP (multidrug resistance protein), a 190,000 integral membrane phosphoglycoprotein that is overexpressed in some drug-selected resistant cell lines [49661]
Propagation: ATCC complete growth medium: The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 20%.
Temperature: 37.0°C
Subculturing: Protocol: Remove medium, and rinse with 0.25% trypsin, 0.03% EDTA solution. Remove the solution and add an additional 1 to 2 ml of trypsin-EDTA solution. Allow the flask to sit at room temperature (or at 37C) until the cells detach. Add fresh culture medium, aspirate and dispense into new culture flasks. Add fresh culture medium, aspirate and dispense into new culture flasks.
Subcultivation Ratio: A subcultivation ratio of 1:3 to 1:6 is recommended
Medium Renewal: 3 to 5 times per week as culture becomes acidic
Preservation: Freeze medium: Complete growth medium 95%; DMSO, 5%
Storage temperature: liquid nitrogen vapor phaseliquid nitrogen vapor phase
Doubling Time: 24 hrs
